چکیده

The paper presents a validated CFD model for the flow in a bench-scale spinner flask that features a centrally-located magnet-driven impeller. Model development found that use of the immersed solid mesh method resulted in better correspondence with observations and μPIV measurements than the more established sliding mesh method in ANSYS-CFX. Analysis of spatial distributions suggested stresses and turbulence in a bench-scale stirredflask bioreactor featuring vertical outer-wall baffles were of a magnitude which could result in inhibited growth due to cell damage if the angular velocity was high enough, even when the magnitude of the Reynolds number indicated that the flow was barely turbulent.

Experimental Characterisation of Fluid Mechanics in a Spinner Flask Bioreactor

The spinner flask bioreactor has been widely used in in vitro cell culturing processes due to its superiority in providing a homogeneous culture environment compared to traditional culturing methods. However, there is limited understanding of the flow fields in these bioreactors, and optimum culture conditions are yet to be determined. This article presents the experimental characterization of ...

Flow Characterization of a Spinner Flask for Induced Pluripotent Stem Cell Culture Application

We present detailed quantitative measurement analyses for flow in a spinner flask with spinning rates between 20 to 45 RPM, utilizing the optical velocimetry measurement technique of Particle Image Velocimetry (PIV). A partial section of the impeller was immersed in the working fluid to reduce the shear forces induced on the cells cultured on microcarriers. Higher rotational speeds improved the...

In vitro culture of primary human myoblasts by using the dextran microcarriers Cytodex3®.

INTRODUCTION Primary cells in vitro culture scale-up is a crucial issue in cell-based tissue and organ regeneration therapy. Reducing costs and space occupied by the cells cultured in vitro has been an important target. Cells cultured in vitro with the use of bioreactor with dextran microcarriers (Cytodex®) have potentially a chance to meet many of the cell therapy requirements. MATERIAL AND ...

Bovine myoblast cell production in a microcarriers-based system

For several tissue engineering applications, in particular food products, scaling up culture of mammalian cells is a necessary task. The prevailing method for large scale cell culture is the stirred tank bioreactor where anchor dependent cells are grown on microcarriers suspended in medium. We use a spinner flask system with cells grown on microcarriers to optimize the growth of bovine myoblast...

Revisiting the determination of hydromechanical stresses encountered by microcarriers in stem cell culture bioreactors

Background Expansion of mesenchymal stem cells (MSC) is one of the key steps for their use in tissue engineering or cell therapies. Today, expansion processes are mainly based on the use of microcarriers to allow large interfacial adherence areas [1]. However, this culture technology is known to be practically limited to low agitation intensity and microcarrier concentrations due to possible ce...
